Yes. R is one of the tools that can be used to create an effective market segmentation analysis. If you have a background in other statistical tools like SPSS, SAS, BMDP, Statistica or SYSTAT, feel free to use those.

No. It would be helpful if you have a background in Statistics but all statistical concepts you will need for market segmentation analysis are explained within this course.
